Acetaldehyde Dehydrogenases in Liver Zonation and Liver Cancer

نویسندگان

چکیده

The liver maintains important homeostatic functions such as metabolism and detoxification. Failure to remove toxic intermediates can cause hepatic damage, fibrosis, even cancer development. This review focuses on acetaldehyde dehydrogenases (ALDHs), a group of key enzymes within the ALDH superfamily with ability convert highly reactive aldehyde substrates corresponding carboxylic acids in NAD(P)-dependent manners. These participate diverse array biological processes detoxification, biosynthesis, antioxidant, regulatory functions. dysfunction disrupt homeostasis, leading buildup, tissue cancer. Here, we examine expression patterns ALDHs adult normal human livers two types cancers—hepatocellular carcinoma cholangiocarcinoma. We also investigated their distributions related zonation. observations provide deep insights into previously unrecognized spatial temporal regulation

Inhibition by ethanol, acetaldehyde and trifluoroethanol of reactions catalysed by yeast and horse liver alcohol dehydrogenases.

1. Produced inhibition by ethanol of the acetaldehyde-NADH reaction, catalysed by the alcohol dehydrogenases from yeast and horse liver, was studied at 25 degrees C and pH 6-9. 2. The results with yeast alcohol dehydrogenase are generally consistent with the preferred-pathway mechanism proposed previously [Dickenson & Dickinson (1975) Biochem. J. 147, 303-311]. The observed hyperbolic inhibitio...

Acetaldehyde adducts in alcoholic liver disease

Chronic alcohol abuse causes liver disease that progresses from simple steatosis through stages of steatohepatitis, fibrosis, cirrhosis, and eventually hepatic failure. In addition, chronic alcoholic liver disease (ALD), with or without cirrhosis, increases risk for hepatocellular carcinoma (HCC). Acetaldehyde, a major toxic metabolite, is one of the principal culprits mediating fibrogenic and ...

Epidemiology and trends in mortality from liver cancer in Iran

Liver cancer is the sixth most common cancer worldwide and the third leading cause of death from cancer. Liver cancer is more prevalent in countries in East Asia, Southeast Asia and the western Central Africa.1 Liver cancer rates in North America and Western Europe, less than 10 cases per hundred thousand people in Asia and Africa between 50-150 cases per hundred thousand people.2 The inc...

Isocitrate Dehydrogenases in Rainbow-trout Liver

1. The kinetics of the thermally induced enzyme variants of the supernatant NADP-isocitrate dehydrogenase from rainbow-trout liver are investigated. 2. Fish acclimatized to 20C (cold-adapted enzyme) and 170C (warm-adapted enzyme) show different relative distributions of the three NADP-isocitrate dehydrogenase isoenzymes; this has been demonstrated with electrophoresis and electrofocusing techni...

Lipid zonation and phospholipid remodeling in nonalcoholic fatty liver disease

Nonalcoholic fatty liver disease (NAFLD) can progress from simple steatosis (i.e., nonalcoholic fatty liver [NAFL]) to nonalcoholic steatohepatitis (NASH), cirrhosis, and cancer. Currently, the driver for this progression is not fully understood; in particular, it is not known how NAFLD and its early progression affects the distribution of lipids in the liver, producing lipotoxicity and inflamm...
